The ribosomal fraction from rat thyroid glands has been isolated after in vivo and in vitro labeling with tritiated leucine by means of differential centrifugation, Na-DOC treatment and ultracentrifugation in sucrose density gradients. The single ribosomes (78 S) have been separated from heavier aggregates (polyribosomes). These last particles appear of at least two classes, one having sedimentation coefficient of about 180 S and the other of 230-250 S. The first class is composed of aggregates of 4-6 ribosomes and the second of 12-15 ribosomes, as shown by electron microscopy. The nascent polypeptide chains labeled with the radioactive aminoacid have been detached from the polyribosomes and examined by density gradient centrifugation and immunoprécipitation. They have sedimentation coefficient less than 4 S and react up to 25% with an antiserum against thyroglobulin isolated from the same animal species. The results indicate that thyroid cells synthesize polypeptide chains whose molecular weight is much smaller than that reported for the subunit of 19 S thyroglobulin.
